Transfer-RNA
Transfer-RNA, tRNA, niedermolekulare, nicht an Proteine gebundene Form der Ribonucleinsäure im Cytoplasma; überträgt Aminosäuren bei der Proteinsynthese. Der Anteil der tRNA an der Gesamt-RNA der Zelle liegt bei 10–15%, die Sedimentationskonstante beträgt etwa 4S. Für jede der 20 proteinogenen Aminosäuren gibt es eine, meist sogar mehrere verschiedene tRNA-Sorten. Insgesamt kennt man ca. 60 Sorten, die sich in ihrer Basensequenz (Primärstruktur) unterscheiden, außerdem noch tRNAs in Chloroplasten und Mitochondrien. Aufgrund von Basenpaarungen besitzt die tRNA eine charakteristische Sekundärstruktur, die einem Kleeblatt ähnelt. Am 5'-Ende des Moleküls befindet sich stets ein Guanosin und am 3'-Ende die Nucleotidsequenz Cytosin-Cytosin-Adenin, an die die Aminosäure gebunden wird. Durch diese »Beladung« entsteht die Aminoacyl-tRNA, die die Aminosäure auf die in Synthese befindliche Polypeptidkette am Ribosom überträgt. An einer der Schleifen ragen drei freie Basen nach außen, das Anticodon, mit dem sich die tRNA über Wasserstoffbrückenbindungen zwischen den komplementären Tripletts an das komplementäre Codon der mRNA anheften kann. Die Aminosäure wird so an der richtigen Position in die wachsende Polypeptidkette eingebaut. Viele Nucleotiden der tRNA enthalten modifizierte
Basen; diese sogenannten »seltenen Basen« induzieren wahrscheinlich die Schleifenbildung, sind aber in der prä-tRNA noch nicht enthalten.
